RESUMO

The N6-methyladenosine (m6A) modification of RNA has been reported to remodel gene expression in response to environmental conditions; however, the biological role of m6A in social insects remains largely unknown. In this study, we explored the role of m6A in the division of labour by worker ants (Solenopsis invicta). We first determined the presence of m6A in RNAs from the brains of worker ants and found that m6A methylation dynamics differed between foragers and nurses. Depletion of m6A methyltransferase or chemical suppression of m6A methylation in foragers resulted in a shift to 'nurse-like' behaviours. Specifically, mRNAs of dopamine receptor 1 (Dop1) and dopamine transporter (DAT) were modified by m6A, and their expression increased dopamine levels to promote the behavioural transition from foragers to nurses. The abundance of Dop1 and DAT mRNAs and their stability were reduced by the inhibition of m6A modification caused by the silencing of Mettl3, suggesting that m6A modification in worker ants modulates dopamine synthesis, which regulates labour division. Collectively, our results provide the first example of the epitranscriptomic regulation of labour division in social insects and implicate m6A regulatory mechanism as a potential novel target for controlling red imported fire ants.

The evolution of insect vector-pathogen relationships has long been of interest in the field of molecular ecology. One system of special relevance, due to its economic impacts, is that between Diaphorina citri and 'Candidatus Liberibacter asiaticus' (CLas), the cause of the severe Asian form of huanglongbing. CLas-positive D. citri are more fecund than their CLas-negative counterparts, boosting opportunities for pathogens to acquire new vector hosts. The molecular mechanism behind this life-history shift remains unclear. Here, we found that CLas promoted ovarian development and increased the expression of the vitellogenin receptor (DcVgR) in ovaries. DcVgR RNAi significantly decreased fecundity and CLas titer in ovaries, extended the preoviposition period, shortened the oviposition period and blocked ovarian development. Given their importance in gene regulation, we explored the role of miRNAs in shaping these phenotypes and their molecular triggers. Our results showed that one miRNA, miR-275, suppressed DcVgR expression by binding to its 3' UTR. Overexpression of miR-275 knocked down DcVgR expression and CLas titer in ovaries, causing reproductive defects that mimicked DcVgR knockdown phenotypes. We focused, further, on roles of the Juvenile Hormone (JH) pathway in shaping the observed fecundity phenotype, given its known impacts on ovarian development. After CLas infection, this pathway was upregulated, thereby increasing DcVgR expression. From these combined results, we conclude that CLas hijacks the JH signalling pathway and miR-275, thereby targeting DcVgR to increase D. citri fecundity. These changes simultaneously increase CLas replication, suggesting a pathogen-vector host mutualism, or a seemingly helpful, but cryptically costly life-history manipulation.

BACKGROUND: The bites and blood sucking of bed bugs (Cimex spp.) (Hemiptera: Cimicidae) pose a serious threat to human physical and mental health. Application of an effective repellent can prevent or reduce bed bug bites. Previous studies on repellent screening mainly focused on Cimex lectularius L. In this study, we investigated the repellent effect of two safe food additives, ethyl anthranilate (EA) and butyl anthranilate (BA), against Cimex hemipterus (F.), and also explored the role of antennae and mouthparts on C. hemipterus perception of repellents. RESULTS: Both EA and BA had a strong repellent effect against tropical bed bugs and their repellency was similar or lower than that of N,N-diethyl-3-methyl benzoyl amide, depending on whether or not a CO2 source was present. EA had higher repellency than BA and exhibited repellency to C. hemipterus for 3 days when a CO2 source was present. C. hemipterus avoided resting on 20% EA- and BA-treated harborages. Applying 20% EA and BA on rabbit skin significantly reduced the blood intake of C. hemipterus within 2 h. C. hemipterus could perceive EA and BA after their antennae or mouthparts or both antennae and mouthparts were removed. CONCLUSION: Both EA and BA had strong repellency against C. hemipterus, with EA being more repellent. Ablation of antennae and mouthparts did not affect the perception of EA and BA by C. hemipterus. © 2023 Society of Chemical Industry.

Huanglongbing, a globally devastating citrus disease, is associated with Candidatus Liberibacter asiaticus (CLas) and is mainly transmitted by Diaphorina citri. Verification of the distribution and dynamics of CLas in D. citri is critical to understanding CLas transmitted by vectors in nature. Here, the distribution and titers of CLas in different sexes and tissues of D. citri adults were investigated by fluorescence in-situ hybridization (FISH) and quantitative real-time PCR (qRT-PCR). Results showed that CLas had widespread distribution in the brain, salivary glands, digestive system, and reproductive system of both females and males, indicating a systemic infection of CLas in D. citri. Moreover, CLas fluorescence intensity and titers were significantly increased in both the digestive system and the female reproductive system with development and there was a marked decreased in both the salivary glands and the male brain, but there was no significant change in the female brain or the male reproductive system. Furthermore, the distribution and dynamics of CLas in embryos and nymphs were investigated. CLas was observed in all laid eggs and subsequent first-second-instar nymphs, indicating that a high percentage of embryos and nymphs resulting from infected D. citri mothers were infected with CLas.

Powdery mildew caused by Blumeria graminis f. sp. tritici is a threat to wheat production in China. Mapping quantitative trait loci (QTL) for resistance to powdery mildew and developing breeder-friendly markers are important initial steps in breeding resistant cultivars. An all-stage resistance gene and several QTL were identified using a population of 254 recombinant inbred lines developed from a Jingdong 8/Aikang 58 cross. The population was evaluated for powdery mildew resistance across six field environments over three consecutive growing seasons utilizing two different mixtures of B. graminis f. sp. tritici isolates, named #Bgt-HB and #Bgt-BJ. Using genotypic data obtained from the Wheat TraitBreed 50K single-nucleotide polymorphism array, seven stable QTL were identified on chromosome arms 1DL, 2AL, 2DS, 4DL, 5AL, 6BL.1, and 6BL.2. The QTL on 2AL conferred all-stage resistance to B. graminis f. sp. tritici race E20 in greenhouse tests and explained up to 52% of the phenotypic variance in field trials but was resistant only against #Bgt-HB. The gene involved in this QTL was predicted to be Pm4a based on genome location and gene sequence. QPmja.caas-1DL, QPmja.caas-4DL, and QPmja.caas-6BL.1 were identified as potentially new QTL for powdery mildew resistance. QPmja.caas-2DS and QPmja.caas-6BL.1 were effective against both B. graminis f. sp. tritici mixtures, indicating their probable broad-spectrum resistance. A Kompetitive allele-specific PCR marker closely linked to QPmja.caas-2DS was developed and validated in a panel of 286 wheat cultivars. Because both Jingdong 8 and Aikang 58 have been leading cultivars and breeding parents, the QTL and marker reported represent valuable resources for wheat researchers and breeders.

Fusarium concentricum Nirenberg & O' Donnell (Ascomycota: Hypocreales) is a fungal species known to infect plants, but never reported as entomopathogenic. Polychrosis cunninhamiacola Liu et Pei (Lepidoptera: Tortricidae: Olethreutinae) is a major and widespread insect pest causing economic losses to cultivated Chinese fir Cunninghamia lanceolata (Lamb.) Hook. It is routinely controlled by extensive use of chemical insecticides, which is perceived as environmentally unsustainable. During March and April of 2019-2020, muscardine cadavers of larvae and pupae of P. cunninhamiacola infected with growing fungus were collected in a fir forest in northern Guangdong Province, China. Conidia were isolated and cultured on PDA medium, from which the fungal strain was identified as F. concentricum FCPC-L01 by morphology and by sequence alignment match with Tef-1α gene. Pathogenicity bioassays at the conidial concentration 1 × 107 revealed P. cunninhamiacola adults and Danaus chrysippus (L.) (Lepidoptera: Nymphalidae) larvae are sensitive to the fungal infection, but not the fire ant Solenopsis invicta Buren (Hymenoptera: Formicidae). We believe results indicate this fungal strain might be applicable against specific target insect pests. As this is the first record of a natural infection caused by F. concentricum in insects, we propose host specificity tests should be done to evaluate its potential as a biocontrol agent.

Numerous bed bug research papers have been published in the past 20 yr as a result of bed bug (Cimex spp.) (Hemiptera: Cimicidae) resurgence in the world. Yet, few of them focused on the management of the tropical bed bug, C. hemipterus (F.). Here, we describe a case of tropical bed bug infestation in two dormitory buildings and effectiveness of a tropical bed bug treatment program. The study site consisted of 125 dormitories in two buildings. An initial building-wide monitoring with ClimbUp interceptors revealed 25 infestations. The spatial distribution of bed bug infested rooms showed a significant aggregated distribution pattern with same infestation status for neighboring units sharing walls. All infested rooms were monitored every 2 wk and treated using a combination of steam and diatomaceous earth (DE) dust application if bed bugs were still found. For the 25 initially identified infested rooms, after 14 wk treatment, 44% of them no longer had bed bugs, and the mean number of bed bugs captured per room decreased by 94.1%. A combination of steam and DE dust treatment is an effective strategy for suppressing tropical bed bug infestations in dormitory environment.

The tropical bed bug, Cimex hemipterus (F.) (Hemiptera: Cimicidae) is an important public-health pest that feeds on the blood of humans and some other animals. To explore the function of the target genes of C. hemipterus, it is essential to select suitable reference genes for the accurate quantification of gene expression. Here, we selected 10 frequently used reference genes in insects and evaluated their stability in C. hemipterus under various biotic (developmental stage, sex, and tissue) and abiotic (gas stimulation and temperature) conditions through RefFinder (which integrates four computational programs: geNorm, NormFinder, BestKeeper, and ∆Ct). Results indicate that the optimal combination of reference genes for each experimental condition was as follows: RPL8 and EF1α for the developmental stage (eggs, early instar nymphs, late instar nymphs, and adults), RPL8 and RPS16 for adult sex, RPL8 and RPL11 for adult tissue (head, thorax, abdomen, and legs), RPL8 and ß-tubulin for gas stimulation (air and carbon dioxide), and RPL8 and NADH for temperature (0, 5, 17, 30, and 37 °C). Finally, the expression pattern of the HSP70 and GR21 genes were analyzed, and the results highlight the importance of appropriate reference-gene selection. Our results provide a comprehensive list of optimal reference genes from C. hemipterus for the first time, which will contribute to accurately analyzing the expression of target genes.

The melon fruit fly, Bactrocera cucurbitae (Coquillett) and the pumpkin fruit fly, Bactrocera tau (Walker) (Diptera: Tephritidae), are important invasive pests on Cucurbitaceous hosts. The acute toxicity of five insecticides to females of these two fruit fly species was bio-assayed by membrane method, and effects of their sublethal concentration on the survival, reproduction, and ovary development of females were evaluated in the laboratory. Results showed that based on the LC50 values, both B. cucurbitae and B. tau were the most susceptible to trichlorfon. After treatment with sublethal concentration (LC30) of trichlorfon, abamectin+ß-cypermethrin, spinetoram, and lambda-cyhalothrin, the female survival days of the two Bactrocera species were significantly shortened compared with the control. Moreover, the fecundity of two Bactrocera species was remarkably decreased, after exposure to abamectin+ß-cypermethrin and trichlorfon LC30. However, the sublethal concentration (LC30) of insecticides had no significant influence on the egg hatchability of the fruit flies. Furthermore, after treatment with abamectin+ß-cypermethrin LC30, the ovary length, width, and egg load of B. cucurbitae were significantly lower than that of the control; however, only the ovarian length and egg load of B. tau were significantly decreased on the 16th and 20th day. In conclusion, abamectin+ß-cypermethrin has an excellent insecticidal activity against B. cucurbitae and B. tau.

Tacrolimus is an important immunosuppressant produced by microbial fermentation. In this study, a modified nanoscale polymeric adsorbent, Ag+-exchanged resin, was prepared and studied for the preparative separation and purification of tacrolimus from fermentation broth of Streptomyces tsukubaensis. The performance and absorption characteristics of the modified nanoscale polymeric adsorbent namely Ag-NPS was evaluated. Notably, Ag-NPS resin displayed the pronounced separation capacities for tacrolimus and its equilibrium adsorption data was well-fitted to the Langmuir isotherm. Moreover, the dynamic adsorption and desorption tests was carried out to obtain the operational parameters for further purification of tacrolimus. Finally, tacrolimus and the two major impurities, ascomycin and dihydrotacrolimus, were separated well in the scale-up purification process. The purity and recovery of tacrolimus was recorded to be 99.12±0.25% and 90.41±2.05%. In conclusion, this method displayed a high potential for separation and purification of tacrolimus and other unsaturated bioactive compounds in high yield from the fermentation broth.

Trichogramma pretiosum Riley is an important natural enemy and biological control agent of lepidopteran pests. Wolbachia is an intracellular endosymbiont that induces parthenogenesis in the parasitoid T. pretiosum. In this paper, the sublethal effects of the antibiotic tetracycline hydrochloride on the development and reproduction of T. pretiosum were studied. Emerged females were fed with sublethal concentrations (LC5, LC15, and LC35) of tetracycline for ten generations. The biological parameters (longevity, parasitized eggs, and fecundity) of treated females significantly reduced compared with the control Moreover, the percentage of female offspring in the treatments significantly reduced, but the percentage of male offspring significantly increased. In addition, the Wolbachia titer sharply reduced after two generations of antibiotic treatments, but it could still be detected even after ten successive generations of antibiotic treatments, which indicated that Wolbachia was not completely removed by sublethal concentrations of tetracycline. The control lines with higher Wolbachia titers produced more female offspring than the tetracycline treatments with lower Wolbachia titers, indicating that the Wolbachia titer affected the sex determination of T. pretiosum. Our results show that sublethal concentrations of tetracycline had adverse effects on the development of T. pretiosum, and Wolbachia titers affected the sexual development of T. pretiosum eggs.

Diaphorina citri is the mainly transmitting vector of the citrus huanglongbing pathogen, which causes severe losses in in the citrus industry. In this study, we isolated a new entomopathogenic fungus, identified as member of Cordyceps fumosorosea based on morphology and ITS sequence analysis. We named C. fumosorosea SCAU-CFDC01 and evaluated its pathogenicity against D. citri nymphs and adults by immersion under laboratory and greenhouse conditions. Results showed that SCAU-CFDC01 was most pathogenic to young nymphs, followed by old nymphs and adults. The LC50 values of the fungus on nymphs and adults showed a declining trend over a 2-7-day period after inoculation. The LT50 (lethal time for a certain concentration to cause 50% mortality) values also presented a decreasing trend along with increasing conidia concentrations. For the results on greenhouse experiments, when 3rd and 5th instar nymphs were inoculated with 1 × 105 conidia mL-1, the survival rate of nymphs were lower, and the emergence rate of adults and female longevity was significantly reduced compared with the control. However, there were no significant effects on sex ratio of adults and male longevity. Our results showed SCAU-CFDC01 was highly pathogenic to D. citri, and may promote mycoparasite development for biological control of D. citri.

Rht-B1b and Rht-D1b, the 'Green Revolution' (GR) genes, greatly improved yield potential of wheat under nitrogen fertilizer application, but reduced coleoptile length, seedling vigor and grain weight. Thus, mining alternative reduced plant height genes without adverse effects is urgently needed. We isolated the causal gene of Rht24 through map-based cloning and characterized its function using transgenic, physiobiochemical and transcriptome assays. We confirmed genetic effects of the dwarfing allele Rht24b with an association analysis and also traced its origin and distribution. Rht24 encodes a gibberellin (GA) 2-oxidase, TaGA2ox-A9. Rht24b conferred higher expression of TaGA2ox-A9 in stems, leading to a reduction of bioactive GA in stems but an elevation in leaves at the jointing stage. Strikingly, Rht24b reduced plant height, but had no yield penalty; it significantly increased nitrogen use efficiency, photosynthetic rate and the expression of related genes. Evolutionary analysis demonstrated that Rht24b first appeared in wild emmer and was detected in more than half of wild emmer and wheat accessions, suggesting that it underwent both natural and artificial selection. These findings uncover an important genetic resource for wheat breeding and also provide clues for dissecting the regulatory mechanisms underlying GA-mediated morphogenesis and yield formation.

Preharvest sprouting (PHS) significantly reduces grain yield and quality. Identification of genetic loci for PHS resistance will facilitate breeding sprouting-resistant wheat cultivars. In this study, we constructed a genetic map comprising 1,702 non-redundant markers in a recombinant inbred line (RIL) population derived from cross Yangxiaomai/Zhongyou9507 using the wheat 15K single-nucleotide polymorphism (SNP) assay. Four quantitative trait loci (QTL) for germination index (GI), a major indicator of PHS, were identified, explaining 4.6-18.5% of the phenotypic variances. Resistance alleles of Qphs.caas-3AL, Qphs.caas-3DL, and Qphs.caas-7BL were from Yangxiaomai, and Zhongyou9507 contributed a resistance allele in Qphs.caas-4AL. No epistatic effects were detected among the QTL, and combined resistance alleles significantly increased PHS resistance. Sequencing and linkage mapping showed that Qphs.caas-3AL and Qphs.caas-3DL corresponded to grain color genes Tamyb10-A and Tamyb10-D, respectively, whereas Qphs.caas-4AL and Qphs.caas-7BL were probably new QTL for PHS. We further developed cost-effective, high-throughput kompetitive allele-specific PCR (KASP) markers tightly linked to Qphs.caas-4AL and Qphs.caas-7BL and validated their association with GI in a test panel of cultivars. The resistance alleles at the Qphs.caas-4AL and Qphs.caas-7BL loci were present in 72.2 and 16.5% cultivars, respectively, suggesting that the former might be subjected to positive selection in wheat breeding. The findings provide not only genetic resources for PHS resistance but also breeding tools for marker-assisted selection.

In order to investigate the effect of Wolbachia on fitness of their hosts, the biological characteristics of a primarily parthenogenetic line and a bisexual restored line (obtained by antibiotic treatment) of Trichogramma pretiosum were compared in the laboratory. Results indicated that both the mean longevity and fecundity of parthenogenetic line (14.2 d and 165 eggs/female) were significantly higher than those in bisexual restored line (8.4 d and 124.2 eggs/female). Both lines of T. pretiosum had the highest daily fecundity (23.3 eggs/female for parthenogenetic line and 19.8 eggs/female for bisexual restored line) on the first day during their reproduction period, and their survival rate and daily fecundity decreased gradually with age; however, the survival rate and daily fecundity of the parthenogenetic line were always higher than those of bisexual restored line, during the entire experimental period. There was no significant difference in emergence rate and deformity rate between the two T. pretiosum lines. The life-table parameter results indicated that net reproduction rate (R0) and mean generation time (T) in parthenogenetic line (133 and 16.8) were significantly higher than those in bisexual restored line (61.7 and 15.5); intrinsic rate of natural increase (rm) and finite rate of increase (λ) in parthenogenetic line (0.29 and 1.34) were higher than those in bisexual restored line (0.26 and 1.3). Based on these results, it can be confirmed that the removal of Wolbachia in the parthenogenetic line had an adverse effect on fitness of T. pretiosum.

Bed bugs (Cimex spp.) (Hemiptera: Cimicidae) are obligatory blood-sucking insects. Research on their behavioral response to host-derived olfactory cues will be helpful for designing more effective management tools. However, previous studies mainly focused on Cimex lectularius L., while the related studies on Cimex hemipterus (F.) have been rarely reported. In this study, we evaluated the behavioral responses of C. hemipterus to one of the crucial host-derived cues - carbon dioxide (CO2), at various concentrations. Pitfall traps baited with CO2 captured significantly more C. hemipterus (72%) than unbaited ones (28%). In the Y-tube test, bed bugs' exploratory activity (response rate) gradually decreased (from 70 to 27%) as the CO2 concentration increased (from 0.9 to 14.5%); and bed bugs exhibited a significant attraction to CO2 when the tested CO2 concentrations were between 0.9 and 7.3%. Cimex hemipterus were no longer attracted to CO2 after their antennae or mouthparts or both antennae and mouthparts were removed. In conclusion, C. hemipterus are attracted to CO2, and both antennae and mouthparts are important for CO2 perception.

OBJECTIVE: This study was conducted to enhance the production of tacrolimus in Streptomyces tsukubaensis by strain mutagenesis and optimization of the fermentation medium. RESULTS: A high tacrolimus producing strain S. tsukubaensis FIM-16-06 was obtained by ultraviolet mutagenesis coupled with atmospheric and room temperature plasma mutagenesis.Then, nine variables were screened using Plackett-Burman experimental design, in which soluble starch, peptone and Tween 80 showed significantly affected tacrolimus production. Further studies were carried out employing central composite design to elucidate the mutual interaction between the variables and to work out optimal fermentation medium composition for tacrolimus production. The optimum fermentation medium was found to contain 61.61 g/L of soluble starch, 20.61 g/L of peptone and 30.79 g/L of Tween 80. In the optimized medium, the production of tacrolimus reached 1293 mg/L in shake-flask culture, and reached 1522 mg/L while the scaled-up fermentation was conducted in a 1000 L fermenter, which was about 3.7 times higher than that in the original medium. CONCLUSIONS: Combining compound mutation with rational medium optimization is an effective approach for improving tacrolimus production, and the optimized fermentation medium could be efficiently used for industrial production.

The entomopathogenic fungus, Cordyceps fumosorosea is a potential eco-friendly biocontrol agent. The present study revealed the entire course of infection of P. xylostella by C. fumosorosea with particular reference to cuticular penetration. Comparative studies on the infection of Plutella xylostella larvae by two strains of C. fumosorosea with different pathogenicity were carried out using light, scanning, and transmission electron microscopy. We found that C. fumosorosea tended to adhere to the cuticle surfaces containing protrusions. Although conidia of the lower pathogenic strain IFCF-D58 germinated, they failed to penetrate and complete the development cycle. In contrast, the higher pathogenic strain IFCF01 began to germinate within 4 h and attached to the cuticle by a thin mucilaginous matrix within 8 h post-inoculation. After 24 h post-inoculation, germ tubes and penetrating hyphae reached the cuticular epidermis and began to enter the haemocoel. Within 36 h post-inoculation, the hyphal bodies colonized the body cavity. Hyphae penetrated from inside to outside of the body after 48 h and sporulated the cadavers. After 72 h post-inoculation, numerous conidia emerged and the mycelial covered the entire cuticular surface. The two strains showed similarities in terms of conidial size and germination rate. However, IFCF-D58 exhibited significantly fewer appressoria and longer penetrating hyphae compared to the more infective IFCF01 on all surface topographies. The current pathogen invasion sequence of events suggested that the aggressive growth and propagation along with rapid and massive in vivo production of blastospores facilitate the conidia of IFCF01 to quickly overcome the diamondback moth's defense mechanism.

From the 1960s to the 1980s, with the implementation of nationwide 'Four-Pest Elimination' campaigns (bed bugs, Cimex lectularius L. and Cimex hemipterus (F.) (Hemiptera: Cimicidae), were listed as one of the targeted pests), bed bug infestations were gradually eliminated in most provinces in China. However, during the last two decades, reports of bed bug infestations in the Pearl River Delta of China have shown an upward trend. Up to now, the bed bug species occurring in this area was much less frequently reported. In this study, we used both morphological and molecular methods to accurately identify the species of bed bugs collected from the cities of Guangzhou and Foshan, China. Results indicated that no significant difference was observed in the mean pronotum width-to-length ratio of Guangzhou (2.6) and Foshan (2.4) specimens; however, both were significantly lower than that of a laboratory strain C. lectularius (3.1). The genetic distances of our specimens with C. hemipterus and C. lectularius were 0-0.2% and 22.2-22.6%, respectively. On the basis of the morphological characteristics and mitochondrial DNA sequence data, it can be affirmed that bed bugs collected from Guangzhou and Foshan were C. hemipterus.

The removal of corpses (aka 'necrophoric behavior') is critical to sanitation in ant colonies. However, little is known about differences in the necrophoric responses of Solenopsis invicta workers towards corpses of nestmates and non-nestmates. We introduced corpses of S. invicta workers from either intracolony (i.e., nestmate) or intercolony (i.e., non-nestmate) origin at the entrance of artificial nests, and recorded workers' aggressive responses and necrophoric behaviors for analysis. Solenopsis invicta workers displayed distinct responses towards corpses of different origins. Specifically, resident workers were more likely to remove fresh non-nestmate corpses than nestmate corpses, but there was no difference regarding corpses that had been dead for 15 min or longer. Resident workers reacted more aggressively to, and removed more quickly, fresh non-nestmate corpses than corpses of their nestmates. On the other hand, there was no significant difference in the removal time between nestmate and non-nestmate corpses that had been dead for 15 min or longer. Resident workers always displayed stronger aggressiveness towards non-nestmate corpses than nestmate corpses, excepting to corpses that had been dead for 6 h, which elicited a response. No significant correlation between the removal times and aggressiveness levels were detected in any treatments. It remains to be tested whether this differential response is adaptive in how it influences colony fitness and competition.
